How to do simple crystal violet staining

Materials:

  • 2 Sterile loop
  • 1 TSA plates
  • 1 Bunsen burner
  • 1 Close pin
  • 2 Slide
  • 1 DI Water
  • 1 Crystal violet
  • 1 Microscope
  • 1 Slide warmer
  • 1 plastic container

To begin simple crystal violet staining, add a drop of DI water on the slides. Then with a sterile loop, loop some of the bacteria on the TSA plate and mix it in with the water on the slides; make sure to spread the liquid to dry it faster and use a different loop for each slide. Now, allow the slides to dry on the slide warmer. When the slides are dry, using a close pin, pass the slide through the fire using a bunsen burner about 3 times quickly. Placed the slides in a plastic container now and drop a few crystal violets stain on them; enough to cover them and wait 1 min. After a min, using DI water, wash off the slides and placed them on a paper towel to dry; lightly damp the top of the slide to dry but not too hard to whip off the bacteria. After the slides are stained purple, look at them under a microscope.
